Environmental surveillance of water sources is important to monitoring viral hepatitis transmission in clinical settings. This study investigated the circulation of hepatitis A (HAV) and E (HEV) viruses in sewage and clinical samples from Argentina. Between 2016 and 2017, 80 raw sewage samples and 86 clinical samples (stool and serum) from suspected cases of hepatitis A and hepatitis E were obtained. HAV and HEV were tested by both real-time and nested PCR. Positive samples were sequenced for genotype determination and phylogenetic analysis. Overall, HAV was recovered in 39% of sewage samples and 61.1% of clinical samples. HEV was detected in 22.5% of sewage samples and 15.9% of clinical samples. HAV was found more frequently in sewage during the winter and in clinical samples in spring; HEV was more prevalent in sewage during summer and in clinical samples in autumn. All HAV isolates belonged to genotype IA and HEV isolates belonged to genotype 3, the most prevalent genotypes in South America. High prevalence of HAV and HEV in environmental and clinical samples in Mendoza, Argentina was observed. These findings reinforce the importance of environmental surveillance and implementation of health strategies to control the spread of HAV and HEV in developing countries.

Despite the long-standing interest in the organization of ant communities across elevational gradients, few studies have incorporated the evolutionary information to understand the historical processes that underlay such patterns. Through the evaluation of phylogenetic α and ß-diversity, we analyzed the structure of leaf-litter ant communities along the Cofre de Perote mountain in Mexico and evaluated whether deterministic- (i.e., habitat filtering, interspecific competition) or stochastic-driven processes (i.e., dispersal limitation) were driving the observed patterns. Lowland and some highland sites showed phylogenetic clustering, whereas intermediate elevations and the highest site presented phylogenetic overdispersion. We infer that strong environmental constraints found at the bottom and the top elevations are favoring closely-related species to prevail at those elevations. Conversely, less stressful climatic conditions at intermediate elevations suggest interspecific interactions are more important in these environments. Total phylogenetic dissimilarity was driven by the turnover component, indicating that the turnover of ant species along the mountain is actually shifts of lineages adapted to particular locations resembling their ancestral niche. The greater phylogenetic dissimilarity between communities was related to greater temperature differences probably due to narrow thermal tolerances inherent to several ant lineages that evolved in more stable conditions. Our results suggest that the interplay between environmental filtering, interspecific competition and habitat specialization plays an important role in the assembly of leaf-litter ant communities along elevational gradients.

Background On encountering a self-inflicted foreign body in the urinary tract, it is common that emergency physicians only consult the department of urology, and no further evaluations from other specialties are sought. Psychological conditions can also involve people with psychiatric disorders who perform self-harming or sexual practices. Many case reports of foreign bodies have been reported in the literature. However, there is little information regarding which specialties to consult in this situation within the emergency department (ED). Methodology This case series study gathered information on 10 cases from patients who attended the ED from 2005 to 2020 with the diagnosis of genital or lower urinary tract foreign body. Results In total, 10 patients were analyzed with a mean age of 37.3 (SD: ±14.1) years. Of the 10 patients, seven (70%) were males, and three (30%) were females. Overall, four (40%) patients presented with lower urinary tract symptoms (dysuria, tenesmus, hematuria, urinary frequency), five (50%) patients had a significant psychiatric history, and eight (80%) patients admitted having these practices for sexual gratification. Conclusions Foreign bodies in the lower urinary tract pose a significant challenge to ED physicians and urologists because some patients do not admit or do not recall inserting foreign bodies. Patients should be interrogated for mental illness, medication use, and a history of foreign bodies in the urinary tract or genitals during the initial evaluation. There is no consensus or screening method for such patients presenting to the ED. Hence, the use of complementary imaging studies and cystoscopy is fundamental for diagnosis. Further, it is essential to perform a psychiatric evaluation to diagnose or address any underlying psychiatric conditions that could cause this behavior.

Biological control of the Mexican fruit fly, Anastrepha ludens, is mainly carried out by releasing parasitoids, such as Diachasmimorpha longicaudata, and by applying entomopathogenic fungi (EPF), such as Metarhizium anisopliae, Beauveria bassiana, or Isaria fumosorosea, which can be applied to the soil or dispersed using infective devices. The combined use of two or more biocontrol agents could improve A. ludens control, but IGP between natural enemies, if it occurs, may have negative effects. We evaluated the effects of EPF on D. longicaudata. First, we determined the susceptibility of adults of D. longicaudata to strains of EPF (Metarhizium robertsii strain V3-160 and M. anisopliae strain MAAP1). We also evaluated the infection of these two fungi on A. ludens larvae parasitized by D. longicaudata. Finally, we determined sub-lethal effects on adults of D. longicaudata that emerged from larvae that had been exposed to low concentrations of M. robertsii. Both fungi caused moderate mortality to D. longicaudata adults. There were no adverse effects on the longevity of parasitoids that emerged from parasitized larvae exposed to M. robertsii. Based on these results, we argue that M. robertsii has the potential to be used for biocontrol of A. ludens, with limited risk to D. longicaudata adults.

The SARS-CoV-2 main protease, also known as 3-chymotrypsin-like protease (3CLpro), is a cysteine protease responsible for the cleavage of viral polyproteins pp1a and pp1ab, at least, at eleven conserved sites, which leads to the formation of mature nonstructural proteins essential for the replication of the virus. Due to its essential role, numerous studies have been conducted so far, which have confirmed 3CLpro as an attractive drug target to combat Covid-19 and have reported a vast number of inhibitors and their co-crystal structures. Despite all the ongoing efforts, D-peptides, which possess key advantages over L-peptides as therapeutic agents, have not been explored as potential drug candidates against 3CLpro. The current work fills this gap by reporting an in silico approach for the discovery of D-peptides capable of inhibiting 3CLpro that involves structure-based virtual screening (SBVS) of an in-house library of D-tripeptides and D-tetrapeptides into the protease active site and subsequent rescoring steps, including Molecular Mechanics Generalized-Born Surface Area (MM-GBSA) free energy calculations and molecular dynamics (MD) simulations. In vitro enzymatic assays conducted for the four top-scoring D-tetrapeptides at 20 µM showed that all of them caused 55-85% inhibition of 3CLpro activity, thus highlighting the suitability of the devised approach. Overall, our results present a promising computational strategy to identify D-peptides capable of inhibiting 3CLpro, with broader application in problems involving protein inhibition.

The arginine repressor (ArgR) regulates the expression of genes involved in arginine biosynthesis. Upon attaining a threshold concentration of arginine in the cytoplasm, the trimeric C-terminal domain of ArgR binds three arginines in a shallow surface cleft and subsequently hexamerizes forming a dimer of trimers containing six Arg co-repressor molecules which are buried at the subunit interfaces. The N-terminal domains of this complex bind to the DNA promoter thereby interrupting the transcription of the genes related to Arg biosynthesis. The crystal structures of the wild type and mutant Pro115Gln ArgR from Corynebacterium pseudotuberculosis determined at 1.7 Å demonstrate that a single amino acid substitution switches co-repressor specificity from Tyr to Arg. Molecular dynamics simulations indicate that the first step, i.e., the binding of the co-repressor, occurs in the trimeric state and that Pro115Gln ArgR preferentially binds Arg. It was also shown that, in Pro115 ArgR hexamers, the concomitant binding of sodium ions shifts selectivity to Tyr. Structural data combined with phylogenetic analyses of ArgR from C. pseudotuberculosis suggest that substitutions in the binding pocket at position 115 may alter its specificity for amino acids and that the length of the protein interdomain linker can provide further functional flexibility. These results support the existence of alternative ArgR regulatory mechanisms in this pathogenic bacterium.

The Mexican fruit fly, Anastrepha ludens Loew, is a significant pest in mango and citrus production areas of Mexico. In this study, we evaluated the effects of some geographic characteristics, rainfall period, soil micro-environmental, and soil coverage variables on the occurrence of entomopathogenic fungi (EPF) associated with A. ludens larvae in soils of mango, grapefruit and mixed crops in central Veracruz state, Mexico. EPF isolates were characterized morphologically and identified by sequence analysis of elongation factor (EF1-1018F, EF1-1620R). We recorded four species of EPF (Metarhizium robertsii J.F. Bisch, S.A. Rehner & Humber [Hypocreales: Cordycipitaceae], M. brunneum Petch [Hypocreales: Cordycipitaceae], M. pinghaense Q.T. Chen & H.L. Guo [Hypocreales: Cordycipitaceae], and Beauveria bassiana (Balsamo) Vuillemin [Hypocreales: Cordycipitaceae]), of which Metarhizium robertsii was the most abundant and the most virulent. Also, we found that rainfall period, organic matter, coverage of herbs and forbs, and calcium levels modulated EPF occurrence. We estimated lethal concentrations for A. ludens larvae of the four most promising isolates, V3-123, V3-160, V1-332, and V3-369. Our results suggest that M. robertsii obtained from agricultural soils holds potential as a biological control agent for A. ludens.

Mexico is one of the most biodiverse countries in the world, with an important proportion of endemism mainly because of the convergence of the Nearctic and Neotropical biogeographic regions, which generate great diversity and species turnover at different spatial scales. However, most of our knowledge of the Mexican ant biota is limited to a few well-studied taxa, and we lack a comprehensive synthesis of ant biodiversity information. For instance, most of the knowledge available in the literature on Mexican ant fauna refers only to species lists by states, or is focused on only a few regions of the country, which prevents the study of several basic and applied aspects of ants, from diversity and distribution to conservation. Our aims in this data paper are therefore (1) to compile all the information available regarding ants across the Mexican territory, and (2) to identify major patterns in the gathered data set and geographic gaps in order to direct future sampling efforts. All records were obtained from raw data, including both unpublished and published information. After exhaustive filtering and updating information and synonyms, we compiled a total of 21,731 records for 887 ant species distributed throughout Mexico from 1894 to 2018. These records were concentrated mainly in the states of Chiapas (n = 6,902, 32.76%) and Veracruz de Ignacio de la Llave (n = 4,329, 19.92%), which together comprise half the records. The subfamily with the highest number of records was Myrmicinae (n = 10,458 records, 48.12%), followed by Formicinae (n = 3,284, 15.11%) and Ponerinae (n = 1,914, 8.8%). Most ant records were collected in the Neotropical region of the country (n = 12,646, 58.19%), followed by the Mexican transition zone (n = 5,237, 24.09%) and the Nearctic region (n = 3,848, 17.72%). Native species comprised 95.46% of the records (n = 20,745). To the best of our knowledge, this is the most complete data set available to date in the literature for the country. We hope that this compilation will encourage researchers to explore different aspects of the population and community research of ants at different spatial scales, and to aid in the establishment of conservation policies and actions. Purpose: The goal was to compare the micronucleus (MN) and dicentric plus ring chromosomes (D + R) assays for dose assessment in cases of partial body irradiations (PBI). Materials and methods: We constructed calibration curves for each assay at doses ranging from 0 to 5 Gy of X-rays at dose rate of 0.275 Gy/min. To simulate partial-body exposures, blood samples from two donors were irradiated with 0.5, 1, 2 and 4 Gy and the ratios of irradiated to unirradiated blood were 25, 50, and 100%. Different tests were used to confirm if all samples were overdispersed or zero-inflated and for partial-body dose assessment we used the Qdr, Dolphin and Bayesian model. Results: In our samples for D + R calibration curve, practically all doses agreed with Poisson assumption, but MN exhibited overdispersed and zero-inflated cellular distributions. The exact Poisson tests and zero-inflated tests demonstrate that virtually all samples of D + R from PBI simulation fit the Poisson distribution and were not zero-inflated, but the MN samples were also overdispersed and zero-inflated. In the partial-body estimation, when Qdr and Dolphin methods were used the D + R results were better than MN, but the doses estimation defined by the Bayesian methodology were more accurate than the classical methods. Conclusions: Dicentric chromosomes continue to prove to be the best biological marker for dose assessment. However exposure scenarios of partial-body estimation, overdispersion and zero-inflation may not occur, it being a critical point not only for dose assessment, but also to confirm partial-body exposure. MN could be used as alternative assay for partial-body dose estimation, but in case of an accident without any information, the MN assay could not define whether the accident was a whole-body irradiation (WBI) or a PBI.

BACKGROUND: Falcipain 2 (FP-2) is the hemoglobin-degrading cysteine protease of Plasmodium falciparum most extensively targeted to develop novel antimalarials. However, no commercial antimalarial drugs based on FP-2 inhibition are available yet due to the low selectivity of most FP-2 inhibitors against the human cysteine proteases. METHODS: A structure-based virtual screening (SVBS) using Maybridge HitFinder™ compound database was conducted to identify potential FP-2 inhibitors. In vitro enzymatic and cell-growth inhibition assays were performed for the top-scoring compounds. Docking, molecular dynamics (MD) simulations and free energy calculations were employed to study the interaction of the best hits with FP-2 and other related enzymes. RESULTS AND CONCLUSIONS: Two hits based on 4-(9H-fluoren-9-yl) piperazin-1-yl) methanone scaffold, HTS07940 and HTS08262, were identified as inhibitors of FP-2 (half-maximal inhibitory concentration (IC50) = 64 µM and 14.7 µM, respectively) without a detectable inhibition against the human off-target cathepsin K (hCatK). HTS07940 and HTS08262 inhibited the growth of the multidrug-resistant P. falciparum strain FCR3 in culture (half-maximal inhibitory concentrations (IC50) = 2.91 µM and 34 µM, respectively) and exhibited only moderate cytotoxicity against HeLa cells (Half-maximal cytotoxic concentration (CC50) = 133 µM and 350 µM, respectively). Free energy calculations reproduced the experimental affinities of the hits for FP-2 and explained the selectivity with respect to hCatK. GENERAL SIGNIFICANCE: To the best of our knowledge, HTS07940 stands among the most selective FP-2 inhibitors identified by SBVS reported so far, displaying moderate antiplasmodial activity and low cytotoxicity against human cells. Hence, this compound constitutes a promising lead for the design of more potent and selective FP-2 inhibitors.

Despite preliminary reports of ants trapped in food-baited fruit fly traps, little is known regarding the identity of the myrmecofauna that can be sampled using this technique. This study aimed to examine the inventory completeness, activity and species occurrence of canopy ant assemblages collected in baited traps used for monitoring fruit flies in different fruit orchards in central Veracruz, Mexico. The trap models used in the sampling were Multilure, McPhail glass, and 500 ml blue polyethylene bottles. Three commercial fruit fly food attractants (CeraTrap, Captor + Borax, and BioLure) and two grape juice products (Jumex grape juice and Tang) were used as baits for sampling. In total 3,626 ant workers belonging to 54 species, 19 genera, 10 tribes, and 5 subfamilies were collected. Among the five food attractants used in this study, CeraTrap recorded a markedly higher inventory completeness, ant activity and species occurrence per trap. This study reports for the first time the use of CeraTrap, as a promising and effective food attractant for collecting the foraging ants in the canopy of agroecosystems, which may be applicable to other habitats such as natural forests, mangroves, or agricultural settings such as coffee plantations.

Riparian vegetation is a distinctive and ecologically important element of landscapes worldwide. However, the relative influence of the surrounding landscape on the conservation of the biodiversity of riparian remnants in human-modified tropical landscapes is poorly understood. We studied the surrounding landscape to evaluate its influence on leaf-litter-ant alpha and beta diversity in riparian remnants in the tropical montane cloud forest region of central Veracruz, Mexico. Sampling was carried out in 12 sites with riparian vegetation during both rainy (2011) and dry (2012) seasons. Ten leaf-litter samples were collected along a 100-m transect per site and processed with Berlese-Tullgren funnels and Winkler sacks. Using remotely-sensed and ground-collected data, we characterized the landscape around each site according to nine land cover types and computed metrics of landscape composition and configuration. We collected a total of 8,684 ant individuals belonging to 53 species, 22 genera, 11 tribes, and 7 subfamilies. Species richness and the diversity of Shannon and Simpson increased significantly in remnants immersed in landscapes with a high percentage of riparian land cover and a low percentage of land covers with areas reforested with Pinus, cattle pastures, and human settlements and infrastructure. The composition of ant assemblages was a function of the percentage of riparian land cover in the landscape. This study found evidence that leaf-litter ants, a highly specialized guild of arthropods, are mainly impacted by landscape composition and the configuration of the focal remnant. Maintaining or improving the surrounding landscape quality of riparian vegetation remnants can stimulate the movement of biodiversity among forest and riparian remnants and foster the provision of ecosystem services by these ecosystems. Effective outcomes may be achieved by considering scientific knowledge during the early stages of riparian policy formulation, in addition to integrating riparian management strategies with broader environmental planning instruments.

Crystallographic data of the dimeric and octameric forms of fragaceatoxin C (FraC) suggested the key role of a small hydrophobic protein-protein interaction surface for actinoporins oligomerization and pore formation in membranes. However, site-directed mutagenesis studies supporting this hypothesis for others actinoporins are still lacking. Here, we demonstrate that disrupting the key hydrophobic interaction between V60 and F163 (FraC numbering scheme) in the oligomerization interface of FraC, equinatoxin II (EqtII), and sticholysin II (StII) impairs the pore formation activity of these proteins. Our results allow for the extension of the importance of FraC protein-protein interactions in the stabilization of the oligomeric intermediates of StII and EqtII pointing out that all of these proteins follow a similar pathway of membrane disruption. These findings support the hybrid pore proposal as the universal model of actinoporins pore formation. Moreover, we reinforce the relevance of dimer formation, which appears to be a functional intermediate in the assembly pathway of some different pore-forming proteins.

Ant-gardens (AGs) are considered one of the most complex mutualist systems between ants and plants, since interactions involving dispersal, protection, and nutrition occur simultaneously in them; however, little is known about the effects of the transformation of ecosystems on their diversity and interactions. In five environments with different land use within an anthropic landscape in southeastern Mexico, we investigated the diversity and composition of epiphytes and host trees of AGs built by Azteca gnava. A total of 10,871 individuals of 26 epiphytic species, associating with 859 AGs located in 161 host trees, were recorded. The diversity and composition of epiphytes tended to be different between environments; however, Aechmea tillandsioides and Codonanthe uleana were the most important species and considered true AG epiphytes, because they were the most frequent, abundant, and occurred exclusively in AGs. Other important species were the orchids Epidendrum flexuosum, Coryanthes picturata, and Epidendrum pachyrachis, and should also be considered true AG epiphytes, because they occurred almost exclusively in the AGs. The AG abundance in agroforestry plantations was similar or even greater than in riparian vegetation (natural habitat). The AGs were registered in 37 host species but were more frequent in Mangifera indica and Citrus sinensis. We conclude that true epiphytes of A. gnava AGs persist in different environments and host trees, and even these AGs could proliferate in agroforestry plantations of anthropic landscapes.

BACKGROUND: Single-dose hepatitis A virus (HAV) vaccination was implemented in all Argentinean children 12 months of age in 2005. Previous studies demonstrated high prevalence of protective antibody response 4 years after single-dose vaccination. This study assessed long-term seroprotection against HAV after vaccination. METHODS: Children who received 1 dose of HAV vaccine at 1 year of age at least 6 years before enrollment were included at 5 centers in Argentina between 2013 and 2014. Demographic and socioeconomic characteristics were collected through a questionnaire. Blood samples were tested for anti-HAV antibodies. Antibody values ≥10 mIU/mL were considered seroprotective. Logistic regression analysis was performed to evaluate the association between demographic and socioeconomic variables and seroprotection. RESULTS: A total of 1088 children were included, with a median postvaccination interval of 7.7 years (range 6.3-9.2 years). Of these children, 97.4% (95% confidence interval: 96.3%-98.3%) had protective antibodies against HAV. No association between demographic or socioeconomic variables and seroprotection was found. Geometric mean concentration of antibody levels against HAV was 170.5 mUI/mL (95% confidence interval: 163.2-178.2 mUI/mL). CONCLUSIONS: Single-dose universal hepatitis A immunization in 1-year-old children resulted in sustained immunologic protection for up to 9 years in Argentina. These findings, along with the low current disease burden, confirm the success of the intervention.

Riparian remnants are linear strips of vegetation immediately adjacent to rivers that may act as refuges for biodiversity, depending on their habitat quality. In this study, we evaluated the role of riparian remnants in contributing to the diversity of leaf-litter ants by determining the relationship between ant diversity and several riparian habitat characteristics within a human-dominated landscape in Veracruz, Mexico. Sampling was carried out in 2012 during both dry and rainy seasons at 12 transects 100 m in length, where 10 leaf-litter samples were collected along each transect and processed with Berlese-Tullgren funnels and Winkler sacks. A total of 8,684 individuals belonging to 53 species, 22 genera, and seven subfamilies were collected. The observed mean alpha diversity accounted for 34.4% of the total species recorded and beta diversity for 65.6%. Species richness and composition were significantly related to litter-layer depth and soil compaction, which could limit the distribution of ant species depending on their nesting, feeding, and foraging habits. Riparian remnants can contribute toward the conservation of ant assemblages and likely other invertebrate communities that are threatened by anthropogenic pressures. In human-dominated landscapes where remnants of riparian vegetation give refuge to a diverse array of myrmecofauna, the protection of the few remaining and well-preserved riparian sites is essential for the long-term maintenance of biodiversity.

INTRODUCTION: Hepatitis E is a disease of global distribution, with significant morbidity and mortality, whose scope and burden continue to emerge in low endemic countries. MATERIAL AND METHODS: In 2012, we studied the prevalence of anti-HEV antibodies in 202 adult serum samples. We also analyzed samples from 143 patients with acute non-A-C hepatitis from January 2011 to December 2013. Acute HEV infections were diagnosed with anti-HEV IgM and/or HEV RNA. HEV RNA was also investigated in 94 swine fecal samples. HEV RNA was sequenced and characterized. RESULTS: We found higher values of prevalence than those previously reported in the 1990s. The overall prevalence of anti-HEV IgG antibodies was 15.4%. The prevalence was 10.6% in the 123 adults voluntarily screened on World Hepatitis Day 2012 in Buenos Aires city and 14.8, 16.7 and 35.7% respectively in the smaller groups of healthcare workers, blood donors and HIV-positive patients from different regions of the country. Nine acute HEV infections were diagnosed in the three years analyzed. We characterized new human variants of subgenotype 3a and 3i. New subgenotype 3i variants were found in swine from two distant provinces closely related to the human ones. CONCLUSIONS: These results enlarge the knowledge of HEV and contribute with new information. However, higher values of prevalence found in small groups need to be confirmed in larger studies. Many aspects of the spectrum of the disease and the reservoirs and routes of transmission are still unknown and thus deserve additional research.
